Enzymatic colorimetry of lecithin and sphingomyelin in aqueous solution.

چکیده
A procedure for the enzymatic determination of lecithin and sphingomyelin in aqueous solution is described. The phospholipids are first dissolved in chloroform:methanol (2:1 by vol), the solvent is evaporated, and the residue is redissolved in an aqueous zwitterionic detergent solution. The enzymatic reaction sequences of both assays involve hydrolysis of the phospholipids to produce choline, which is then oxidized to betaine, thus generating hydrogen peroxide. The hydrogen peroxide is subsequently utilized in the enzymatic coupling of 4-aminoantipyrine and sodium 2-hydroxy-3,5-dichlorobenzenesulfonate, an intensely red color being formed. The presence of a non-reacting phospholipid enhances the hydrolysis of the reacting phospholipid. Thus we added lecithin to the sphingomyelin standards and sphingomyelin to the lecithin standards. This precise procedure may be applicable to determination of lecithin and sphingomyelin in amniotic fluid.

The metabolism of sphingomyelin. II. Evidence of an enzymatic deficiency in Niemann-Pick diseae.
The accumulation of excessive quantities of sphingomyelin in tissues of patients with Niemann-Pick disease was demonstrated by Klenk in 19341, 2 and has been amply confirmed by other investigators.3-5 A study by Crocker and Mays6 indicated that the rate of biosynthesis of sphingomyelin in tissues from these patients appeared to be essentially normal. These findings suggested that the metabolic ...

We describe methods for automated enzymatic measurement of lecithin, sphingomyelin, and phosphatidylglycerol in amniotic fluid. Phospholipase C (EC 3.1.4.3) and sphingomyelin phosphodiesterase (EC 3.1.4.12) are reacted with lecithin and sphingomyelin, respectively, to liberate phosphocholine. Phosphocholine is then reacted with alkaline phosphatase, choline oxidase, peroxidase, and 4-aminoantip...

The role of cytidine coenzymes in phospholipide biosynthesis is now well established (1, 2). Since sphingomyelin has a phosphorylcholine (PC) moiety similar to that found in lecithin, it ha.s been proposed (3) that it is synthesized enzymatically in a reaction in which the phosphorylcholine moiety of CDPcholine is transferred to the free primary hydroxyl group of a ceramide (N-acylsphingosine) ...

The relation between the choline containing surfactant phospholipids lecithin and sphingomyelin in amniotic fluid and fetal lung maturity is well established. An enzymatic method that had been automated and optimised for use on a centrifugal analyser was used to measure the total choline containing phospholipids in amniotic fluid. The total time taken for this assay was 10 minutes. The results ...
